Emily Annabeth Locke is the biological mother of John Locke. She was born on October 15, 1940.

As a teenager
At the age of sixteen, Emily was hit by a car while she was six months pregnant with John. While she was being rushed into the hospital, she revealed to the doctors that she was pregnant. She soon gave birth to John, who had to be kept in isolation for the sake of his health. After he regained his health, the nurses were going to allow Emily to hold him for the first time, but she decided she couldn't do it and ran out of the hospital. Emily's mother then asked the nurse about the possibility of adoption, while Richard Alpert looked on. ("Cabin Fever")
Reuniting with Locke
About ten years prior to the crash of Flight 815, Anthony Cooper used Emily as part of a con to get Locke to donate his kidney to him, after his own had failed. During the ploy, she told her son that he was very special, and that he had no father because he was "immaculately conceived". Emily would later reveal Cooper's betrayal, apologizing and telling Locke that she participated in the con for money.("Deus Ex Machina")
Emily was admitted several times to the Santa Rosa Mental Health Institute for a treatable type of schizophrenia. She is addicted to drugs; however, it is unknown if the addiction is related to her treatment. ("Deus Ex Machina")
